研究主題:利用功能性體學研究探討綠豆之豆象抗性與沉香高價值代謝物之研究

       本研究室之主要研究興趣是將功能性體學研究方法應用於轉譯農學研究上,目前著重於
綠豆抗豆象與沉香代謝之研究。

綠豆抗豆象之轉譯農學研究
       綠豆是南亞與東南亞的重要作物,但其貯藏過程會受到豆象蛀蝕並造成經濟上的嚴重損
失。研究室針對不同豆象抗性綠豆進行基因體、轉錄體、蛋白質體與代謝體學研究,已完成
豆象抗性綠豆RIL59 之基因體組裝並整合綠豆轉錄體資料得到4 萬餘筆綠豆基因的註釋。
結合數量遺傳位點研究,於第五條染色體上發現一個與豆象抗性相關的主要遺傳位點( Fig. 1)。
比較基因體、轉錄體與蛋白質體的研究亦顯示蛋白質水解酶抑制劑、BURP-domain蛋白與跳
躍基因可能參與綠豆抵禦豆象的蛀蝕;這些抗豆象候選基因在綠豆抗豆象上所扮演的功能仍
持續釐清中。

沉香樹及沉香香氣成分之研究
       本實驗室亦致力沉香二次代謝物及香氣成分生合成機制、香氣成分差異分析,以及沉
香樹基因體的研究。本研究室先後完成沉香樹(Aquilaria agallocha) 的全基因體定序及基因表現
的分析(Chen et al. BMC Genomics 2014;Kuo etal. BMC Plant Biology 2015) 。最近我們利用懸浮
細胞技術、次世代定序技術及GC-MS ,進行沉香懸浮細胞生合成倍半類香氣成分的生合成
機制的研究。以LC-MS 、GC-MS 進行各種類沉香香氣成份的差異分析(Fig. 2) ,並且利用DNA
序列鑑定沉香之來源樹種,同時整合沉香的化學成分和沉香樹樹種的相關性。
